Acute Phase Protein RID Kits

In addition to ELISA kits for measurement of acute phase proteins we now manufacture radial immunodiffusion (RID) kits for quantitative determination of selected acute phase proteins. The RID technique is extremely simple and elegant, requiring no expensive test equipment. It is particularly suited to the measurement of proteins with serum concentrations of 0.1 mg/ml or higher.

Diluted serum or plasma samples (5 or 10 ml) are placed in wells of an agar plate containing antiserum specific for the test protein (antigen). As the sample diffuses radially from the well a precipitin reaction occurs between the antigen and the specific antibodies, resulting in the formation of a visible precipitin ring, the diameter of which is measured 24-48 hours after sample addition. The diameter of the precipitin ring is proportional to the concentration of antigen in the sample. Antigen concentration is determined by comparison to reference standards.

Please note: Our RID kits are offered with or without magnifiers that allow accurate measurement of the precipitin ring diameter (an illustration of the magnifier is shown below). If you are ordering multiple kits, you need order only one kit with a magnifier.

Serum or plasma samples are diluted 10-100 fold with sample diluent (provided)
Standards are prepared as described in the product insert
Diluted samples & standards (5 or 10 ml) are added to the wells and incubated for 1-2 days at 37 degrees celsius
The diameter of the precipitin rings is determined using the included magnifier/graticule
Analyte concentration is determined by comparison to a standard curve (concentration versus the squared value of the ring diameter)
