The ranges reported for the various kits are those obtained using the sample dilution factor suggested in the kit insert. In most cases, the working ranges of the kits may be extended simply by modifying the sample dilution factor.

General Assay Format (Animal APP ELISA's)
Serum or plasma samples are diluted 1,000 - 100,000 fold with sample diluent (provided)
Standards are prepared as described in the product insert
Diluted samples & standards (100 ml) are added to the wells and incubated for 30 - 45 minutes
The wells are washed with water or wash buffer
HRP conjugated detection antibody (100 ml) is added to the wells and incubated for 30- 60 minutes
The wells are washed
HRP substrate (100 ml) is added
After 20 minutes the reaction is quenched by addition of Stop Solution (100ml)
Absorbance is read at 405 or 450 nm
Analyte concentration is determined by comparison to a standard curve

Assay Format (Human CRP ELISA)
Serum or plasma samples are diluted 100 fold with sample diluent (provided)
Standards are prepared as described in the product insert
Diluted samples and standards (10 ml) are added to microtiter wells
HRP conjugated detection antibody (100 ml) is added
The plate is incubated for 45 minutes, then washed with distilled or deionized water
HRP substrate (100 ml) is added
After 20 minutes the reaction is quenched by addition of Stop Solution (100 ml)
Absorbance is read at 450 nm
Analyte concentration is determined by comparison to a standard curve

The Figure below illustrates the levels of four acute phase proteins (a-1-acid glycoprotein, ceruloplasmin, CRP and haptoglobin) determined in serum from normal and diseased (adjuvant induced arthritis) rats using ELISA kits manufactured by Life Diagnostics, Inc.
